精神依赖是药物成瘾的核心症状,戒断形成的负性情绪记忆是精神依赖主要原因,也是复吸的重要诱因。以戒断负性情绪为研究重点,聚焦于戒断记忆的突触可塑性研究,有助于对阿片成瘾神经机制的理解,推动从实验室到临床的成果转化。
通过吗啡急性戒断消退学习模型和核团损毁方式,发现腹内侧前额皮层对戒断消退记忆的巩固至关重要。局部注射NMDA受体激动剂D-cycloserine或抑制剂AP5,不仅显著增加或降低ERK/CREB磷酸化水平,也相应增强或减弱戒断记忆的消退。通过染色质免疫沉淀(chromatin immunoprecipitation,ChIP)实验,表明消退训练经ERK/CREB信号通路增加组蛋白H3和H4乙酰化水平,促进BDNF外显子exon I表达。利用细胞膜表面受体蛋白交联实验发现,消退训练降低腹内侧前额皮层细胞膜上GABAA受体水平,增加细胞内受体含量;其分子作用机制为,BDNF经TrkB受体激活Rac1/PAK1 /Cofilin信号通路,重组细胞骨架,转运Arc蛋白至突触后膜,组成复合物促进GABAA受体向膜内转运。通过膜片钳和行为学实验进一步发现,消退训练促进LTP的生成,GABAA受体膜上表达降低是促使LTP生成的重要分子机制。
